Nucleic acid delivery has expanded from recombinant gene expression to a diversity of other applications, such as knockdown with siRNA and genome editing with CRISPR RNPs, to name a few. With these advances in applications, three approaches have been a mainstay for delivering nucleic acid cargo into mammalian and insect cells: chemical transfection, viral transduction, and electroporation. Here, we highlight the primary benefits associated with each method to identify the best approach for a given application and the available resources of the laboratory. No single delivery method is ideal for all situations. Despite this, researchers may routinely employ a suboptimal approach for the sake of familiarity and to avoid startup costs associated with trying a new method. In addition to describing these methods, we also share information about Mirus Bio TransIT® transfection reagents and the Ingenio® EZporator® Electroporation System, which are both easy-to-use and cost-effective.

Dr. Sandy Tseng


Dr. Tseng is a Tech Support Scientist at Mirus. She has a PhD in Biochemistry from the University of Wisconsin-Madison, where she studied regulation of eukaryotic transcription with a variety of approaches ranging from transcriptomics to kinase chemical-genetic inhibition. Her versatility and mechanistic understanding of gene expression is applied to supporting customers that use Mirus transfection and electroporation reagents.
